Aptamers constitute an emerging class of molecules designed and selected to recognize any given target that ranges from small compounds to large biomolecules, and even cells. However, the underlying physicochemical principles that govern the ligand‐binding process still have to be clarified. A major issue when dealing with short oligonucleotides is their intrinsic flexibility that renders their active conformation highly sensitive to experimental conditions. To overcome this problem and determine the best experimental parameters, an approach based on the design‐of‐experiments methodology has been developed. Here, the focus is on DNA aptamers that possess high specificity and affinity for small molecules, L ‐tyrosinamide, and adenosine monophosphate. Factors such as buffer, pH value, ionic strength, Mg2+‐ion concentration, and ligand/aptamer ratio have been considered to find the optimal experimental conditions. It was then possible to gain new insight into the conformational features of the two ligands by using ligand‐observed NMR spectroscopic techniques and molecular mechanics.  相似文献

Vicinal diketones, namely diacetyl (DC) and pentanedione (PN), are compounds naturally found in beer that play a key role in the definition of its aroma. In lager beer, they are responsible for off-flavors (buttery flavor) and therefore their presence and quantification is of paramount importance to beer producers. Aiming at developing an accurate quantitative monitoring scheme to follow these off-flavor compounds during beer production and in the final product, the head space solid-phase microextraction (HS-SPME) analytical procedure was tuned through experiments planned in an optimal way and the final settings were fully validated. Optimal design of experiments (O-DOE) is a computational, statistically-oriented approach for designing experiences that are most informative according to a well-defined criterion. This methodology was applied for HS-SPME optimization, leading to the following optimal extraction conditions for the quantification of VDK: use a CAR/PDMS fiber, 5 ml of samples in 20 ml vial, 5 min of pre-incubation time followed by 25 min of extraction at 30 °C, with agitation. The validation of the final analytical methodology was performed using a matrix-matched calibration, in order to minimize matrix effects. The following key features were obtained: linearity (R2 > 0.999, both for diacetyl and 2,3-pentanedione), high sensitivity (LOD of 0.92 μg L−1 and 2.80 μg L−1, and LOQ of 3.30 μg L−1 and 10.01 μg L−1, for diacetyl and 2,3-pentanedione, respectively), recoveries of approximately 100% and suitable precision (repeatability and reproducibility lower than 3% and 7.5%, respectively). The applicability of the methodology was fully confirmed through an independent analysis of several beer samples, with analyte concentrations ranging from 4 to 200 g L−1.  相似文献

Experiments were carried out using a strong acid cation exchange resin with a particle size of 75–150 μm, termed as “fine resin” in hydrogen ion form for the elution of individual lanthanides Sm, Eu, Gd, Tb, and Dy that are produced as fission products in the spent nuclear fuel and generated in the effluent during reprocessing of spent nuclear fuel. Batch experiments were carried out to study the effect of concentration of nitric acid on distribution coefficient. The distribution coefficient values for these individual lanthanides were determined in nitric acid medium in the concentration range of 0.01–4.0 N. Uptake of each individual lanthanide by resin was increased with increased nitric acid concentration from 0.01 to 0.5 N and remained similar from 0.5 to 1.0 N and decreased thereafter up to 4.0 N. Column experiments were also carried out using the same resin to study the parameters like pH of the eluent, flow rate, and resin bed height under isocratic elution conditions for eluting lanthanide elements using α‐hydroxyisobutyric acid as eluent. The results of this study have indicated the possibility for the elution of individual lanthanides.  相似文献

In this article, the authors present finite element analysis and approximation of Burgers’‐Fisher equation. Existence and uniqueness of weak solution is proved by Galerkin's finite element method for non‐smooth initial data. Next, a priori error estimates of semi‐discrete solution in norm, are derived and the convergence of semi‐discrete solution is established. Then, fully discretization of the problem is done with the help of Euler's backward method. The nonlinearity is removed by lagging it to previous known level. The scheme is found to be convergent. Positivity of fully discrete solution is discussed, and bounds on time step are discovered for which the solution preserves its positivity. Finally, numerical experiments are performed on some examples to demonstrate the effectiveness of the scheme. The proposed scheme found to be fast, easy and accurate.© 2017 Wiley Periodicals, Inc. Volatile methylsiloxanes (VMSs) constitute a group of compounds used in a great variety of products, particularly personal care products. Due to their massive use, they are continually discharged into wastewater treatment plants and are increasingly being detected in wastewater and in the environment at low concentrations. The aim of this work was to develop and validate a fast and reliable methodology to screen seven VMSs in water samples, by headspace solid-phase microextraction (HS-SPME) followed by gas chromatography with flame ionization detection (GC-FID). The influence of several factors affecting the extraction efficiency was investigated using a design of experiments approach. The main factors were selected (fiber type, sample volume, ionic strength, extraction and desorption time, extraction and desorption temperature) and optimized, employing a central composite design. The optimal conditions were: 65 µm PDMS/Divinylbenzene fiber, 10 mL sample, 19.5% NaCl, 39 min extraction time, 10 min desorption time, and 33 °C and 240 °C as extraction and desorption temperature, respectively. The methodology was successfully validated, showing low detection limits (up to 24 ng/L), good precision (relative standard deviations below 15%), and accuracy ranging from 62% to 104% in wastewater, tap, and river water samples.  相似文献

The present paper describes results of plate-impact pressure-shear friction experiments conducted to study time-resolved growth of molten metal films during dry metal-on-metal slip under extreme interfacial conditions. By employing tribo-pairs comprising hard tool-steel against relatively low melt-point metals such as 7075-T6 aluminum alloys, interfacial friction stress ranging from 100 to and slip speeds of approximately have been generated. These relatively high levels of friction stress combined with high slip-speeds generate conditions conducive for interfacial temperatures to approach the melting point of the lower melt point metal (Al alloy) comprising the tribo-pair.A Lagrangian finite element code is developed to understand the evolution of the thermo-mechanical fields and their relationship to the observed slip response. The code accounts for dynamic effects, heat conduction, contact with friction, and full thermo-mechanical coupling. At temperatures below the melting point the material is described as an isotropic thermally softening elastic-viscoplastic solid. For material elements with temperatures in excess of the melt point a purely Newtonian fluid constitutive model is employed.The results of the hybrid experimental-computational study provides new insights into the thermoelastic-plastic interactions during high speed metal-on-metal slip under extreme interfacial conditions. During the early part of frictional slip the coefficient of kinetic friction is observed to decrease with increasing slip velocity. During the later part transition in interfacial slip occurs from dry metal-on-metal sliding to the formation of molten Al films at the tribo-pair interface. Under these conditions the interfacial resistance approaches the shear strength of the molten aluminum alloy under normal pressures of approximately 1- and shear strain rates of . The results of the study indicate that under these extreme conditions molten aluminum films maintain a shearing resistance as high as .Scanning electron microscopy of the slip surfaces reveal molten aluminum to be smeared on the tribo-pair interface. Knoop hardness measurements in 7075-T6 Al alloy at various depths from the slip interface indicate that the hardness increases approximately linearly with depth and reaches a plateau at approximately from the surface.  相似文献

The linear viscoelastic behavior of a soda-lime-silica glass under low frequency shear loading is investigated in the glass transition range. Using the time-temperature superposition technique, the master curves of the shear dynamic relaxation moduli are obtained at a reference temperature of 566°C. A method to determine the viscoelastic constants from dynamic relaxation moduli is proposed. However, some viscoelastic constants cannot be directly measured from the experimental curves and others cannot be precisely obtained due to non-linearity effects at very low frequencies. The generalized Maxwell model is investigated from the experimental dynamic moduli without fixing the viscoelastic constants. A set of parameters is shown to be in good agreement with the experimental dynamic relaxation moduli, but does not give the correct values of the viscoelastic constants of the investigated glass. The soda-lime-silica glass exhibits a non-linear viscoelastic behavior at very low stress level which is usually observed for organic glasses. This non-linear behavior is questioned.  相似文献

在容积为0.00814m3的柱形泄爆容器中,对泄爆现象进行实验研究. 容器内充满当量比为1的甲烷-空气预混气,采用底端中心点火,泄爆压力为230±15kPa. 基于k-ε湍流模型和EBU燃烧模型,利用同位网格的SIMPLE算法,对该现象进行了数值模拟. 实验和计算获得的外轴线上4个测压点的压力曲线和外流场的阴影和数值照片,形象地描述了高压泄爆时外部流场的变化. 数值结果与实验结果基本一致. 根据实验和数值结果,详细地讨论了泄爆过程中二次爆炸产生的动力学机理. 泄爆的初始阶段,在破膜激波的引导下,泄出的未燃气体因欠膨胀在外流场形成稀疏波低压区和悬激波高压区. 高压区可燃气体密度和温度上升,成为高密度的预热区域. 随后,火焰以射流形式从泄爆口泄出,点燃可燃气云. 受湍流等因素的影响,特别在高密度的预热区域,燃烧速率可能迅速增大,从而导致二次爆炸.  相似文献
